Effects of guggulsterone derivatives GG-50B and GG5 on IκBα phosphorylation and IKK activity in COLO 205 cells stimulated with TNF-α. COLO 205 cells were pretreated with the guggulsterone derivatives GG-50B and GG-52 (10 or 50 μM) for 24 h and then stimulated with TNF-α (10 ng/ml) for 30 min. (a) Immnoblot analysis for phospho-IκBα, IκBα and actin was performed. The data are representative of more than three independent experiments. (b) IKK kinase activity was measured using the HTScan IKK-β kinase assay kit. Data are expressed as mean-fold induction ±s.e.m. of kinase activity relative to untreated controls (n=5). *P<0.05 compared with TNF-α alone.
