Mechanism of cysteinyl leukotriene (Cys-LT) secretion in acute cholestasis and normal liver. Data are expressed as the mean±s.e.m. (a) The infusion of Zymosan (Zy, 150 μg/ml, n=6) from 40 to 46 min after starting liver perfusion increased the sinusoidal Cys-LT efflux (pg/min × g liver) compared with the control group that did not undergo Zymosan infusion (#P<0.05) in BDL animals. Additionally maximal Cys-LT efflux was significantly (*P<0.05) higher in BDL animals than in sham-operated animals. (b) In sham-operated animals, the biliary Cys-LT efflux significantly (*P<0.05) increased after KC activation with Zymosan (150 μg/ml, 40–46 min, n=6) compared with the controls. 5-LO inhibition with MK-886 (3 μM, 30–60 min, n=6) almost completely abolished biliary Cys-LT efflux following KC activation (**P<0.05).
