In vitro colony formation. When a total of 5 × 104 cells were plated in methylcellulose-containing medium, colonies were obtained from CD20− CD138− subpopulation but not from CD20+ CD138− and CD20+ CD138+ subpopulations. When a total of 1 × 105 cells were plated, the number of colonies obtained from CD20− CD138− subpopulation was significantly higher than that of CD20+ CD138+ and CD20+ CD138− subpopulations. The number of colonies was counted on day 23 when a total of 1 × 105 cells were plated, and on day 35 when a total of 5 × 104 cells were plated. *P<0.05 when compared with CD20− CD138− cells.
