From: miR-101a and miR-30b contribute to inflammatory cytokine-mediated β-cell dysfunction

miR-101a binds to Onecut2 3′-UTR and regulates its expression, but miR-30b does not. (a) Potential binding sites of miR-101a and miR-30b within the Onecut2 3′-UTR are shown. HEK293 cells were transiently co-transfected with relative luciferase reporter constructs containing either a wild-type (WT) or a mutated (MUT) Onecut2-binding site sequences with miR-101a or miR-30b mimics. Two days later, luciferase activities were measured and normalized to a renilla luciferase activity. The data shown are the mean±s.d. of three replicates and are representative of three independent experiments. (b) Only miR-101a but not miR-30b downregulates Onecut2 expression. A real-time PCR analysis was performed 36 h after transfection with miR-101a or miR-30b mimics, or with a negative control.
