Src is a common node downstream of ALK and EGFR signaling pathways in NSCLC ALK-positive cells. (a) H2228 and H3122 cells were treated with CEP-28122 and with 5 μM SKI-I (S) in 0.05% FCS for 24 h and for 12 h, respectively. Total cell lysates were blotted with the indicated antibodies. (b–e) H2228 and H3122 cells were treated with CEP-28122 alone or in combination with 5 μM Src kinase inhibitor-1 (SKI-1), and with or without EGF stimulation (10 ng/ml) every 24 h. The cell proliferation rate was evaluated with the Cell Titer-Glo Luminescent cell viability assay (b, d) and the percentage of apoptotic cells was quantified by tetrametylrodamine methyl ester staining followed by fluorescence-activated cell sorting analysis (c, e).
