AII is involved in the control of neonatal renal hemodynamics and has been shown to interact with nitric oxide (NO) according to a specific developmental pattern. Distribution of AII receptor AT1 and AT2 subtypes in the kidney is developmentally regulated. The present study examines the effect of AT1 and AT2 receptors specific antagonists on AII/NO interactions in the in vitro, developing rabbit isolated perfused kidney (IPK).

Methods: Experiments were conducted on rabbit IPK provening form 7 and 42 day (d)-old animals. Kidneys were perfused with an oxygenated, modified Krebs-Enselheit medium at constant flow, in the presence of indomethacin 10-5M. Changes in perfusion pressure were measured in response to AII, 10-10M and 10-8M, in the presence of L158809 (L), 0.5 μM, or PD123319 (PD), 0.5 μM, respectively AT1- and AT2-receptors antagonists, in control conditions and in the presence of L-NAME, a NO-synthase inhibitor, 10-4M.
