Effects of MITF and its mutants on TYR promoter activity. The luciferase reporter plasmid TYR-Luc was transiently transfected into 293T cells (a) or melanoma UACC903 cells (b) in combination with WT or mutant MITF expression vectors. The basal level of luciferase was set as 1. Data from all other transfections are presented as fold induction above this level. Luciferase activity was normalized to β-galactosidase activity. Each value shown is the mean ± SD of three replicates from a single assay. The results shown are representative of at least three independent experiments (**p < 0.01,***p < 0.001 compared with the basal activity, ###p < 0.001 compared with the WT MITF, unpaired Student’s t-test)
