Fig. 1: Experimental timeline and correlational analyses.

a Rats underwent testing for stress- and anxiety-related behaviors (OFT: open field test; EPM: elevated-plus maze) and nociceptive threshold (TF: tail flick) prior to jugular catheterization surgery, with tests repeated following heroin experience. Heroin self-administration occurred for 3 weeks (12-h/session, 4 days a week) followed by a progressive ratio test and an additional 3 days of self-administration training. Next, rats underwent a within-session extinction-heroin prime reinstatement test (6-h). Extinction training sessions (2-h/session) then occurred followed by a cue-induced reinstatement test (2-h). Spleen sample were collected for genetic analyses at the conclusion of training. b Correlation matrix for behavioral trait covariance. Behavioral testing that occurred prior to heroin experience is designated with a 1, and testing following heroin training as 2. Traits in bold are those with at least one significant QTL identified in GWAS analysis (n = 874).
