The deficiency of FXR results in increased Thoc6 and RUNX2 is ubiquitinated excessively by Thoc6, followed by lysosomal degradation, consequently suppressing osteoblast differentiation leading to osteoporosis; During FXR sufficient, FXR promotes osteoblast differentiation by suppressing Thoc6 expression and reducing ubiquitination of RUNX2 to help maintain healthy bone homeostasis
