Rps27l inactivation in the Trp53+/− background triggers Mdm2 accumulation, which on one hand increases p53 by degrading Mdm4, leading to reduced ligase activity toward p53, and on the other hand binds to Nbs1 to reduce its binding and MRN activation of Atm. Increased p53 induces apoptosis, whereas reduced Atm activation impairs DNA damage response, leading to enhanced sensitivity to radiation
