a Zfp90 interacts with Snf2l via yeast two-hybrid screen. Yeast strain AH109 was co-transfected with Gal4 DNA-binding domain (BD)-fused Zfp90 and Gal4 activating domain (AD)-fused Snf2l. Interaction of BD-p53 and AD-large T antigen was used as a positive control. b Interaction of Zfp90 with the NURF complex was confirmed by co-IP assay. Flag-Bptf, Myc-Zfp90, HA-Snf2l, and His-Rbbp4 were co-transfected into 293T cells for 36 h. β-actin was used to indicate the amount of loading proteins. c BM cell lysates were incubated with anti-Zfp90 for immunoprecipitation assay. β-actin was used to indicate the amount of loading proteins. d BM HSCs from wild-type mice were sorted, fixed and co-stained with the indicated antibodies. Zfp90, red; Snf2l, green; nucleus, blue. Scale bar=2 µm. e GST-tagged Zfp90 was incubated with His-tagged Snf2l, Rbbp4 or Flag tagged Bptf at 4 °C for 4 h, followed by incubation with GST beads and western blot (WB) analysis. β-actin was used to indicate the amount of loading proteins. All data are representative of three independent experiments
