a, b MV4–11 and KG-1α cells were exposed to the indicated concentrations of TPL ± ABT-199 for 48 h, after which western blot analysis was performed to monitor cleavage caspase 3, caspase 9, and PARP. c, d Alternatively, cells were treated with the indicated concentrations of TPL ± ABT-199 for 48 h, after which flow cytometry was performed to assess depolarization of mitochondrial membrane after JC-1 staining. e, f MV4–11 and KG-1α cells were incubated for 24 h with the indicated concentrations of TPL ± ABT-199 in the absence or presence of the pan-caspase inhibitor Z-VAD-fmk, after which the percentage of apoptotic cells was determined by flow cytometry after Annexin-V and PI staining.
