a Cellular extracts from MCF-7 cells stably expressing FLAG-SET7/9 or the vector alone were immunopurified with an anti-FLAG affinity column and eluted using FLAG peptides. The eluates were resolved by SDS–PAGE and analyzed by mass spectrometry. b The column-bound proteins were analyzed by western blot using antibodies against TRIM21. c Co-immunoprecipitation analysis was performed in MCF-7 cells, T-47D cells, MDA-MB-231 cells, and MCF-10A cells with antibodies against SET7/9. Immunocomplexes were then immunoblotted using antibodies against TRIM21, and the reciprocal procedure was also performed. d The GST pull-down assay was performed with GST-SET7/9 or its deletion mutants and in vitro-transcribed and translated full-length TRIM21.
