A THP-1 cells were cultured with rhMANF (2 μg/mL) for the indicated times. Activation of various signaling pathways was measured by western blotting. B THP-1 cells were pretreated with various inhibitors for 1 h, followed by rhMANF (2 μg/mL) treatment for 5 h. Il10 mRNA expression in THP-1 cells was measured by RT-qPCR. C, D THP-1 cells were cultured with p38 inhibitor for 1 h, followed by rhMANF (2 μg/mL) with or without IL-10 (20 ng/mL) treatment for 5 h.
