By cross-linking with FcγRIIIA on MDSCs, immunocomplex activates the cytoplasmic ITAM, which induces the activation of mTOR to promote the expression of transcriptional factor HIF-1α. Furthermore, HIF-1α promote the expression of glycolysis-associated Glut1, HK2 and LDHA to enhance glycolysis, which results in inflammatory cytokines up-regulation leading to the decreased Th1/Th2 ratio and increased Th17/Treg ratio.
