A 143B and HOS cells coexpressed HA-ubiquitin, Flag-EPHA2 and Myc-USP3. EPHA2 ubiquitination was then analyzed by WB. B 143B and HOS cells transfected with control (sh-NC) or USP3 knockdown (sh-USP3) plasmids were immunoprecipitated, and EPHA2 ubiquitination was then detected by Western blotting. C HEK293T cells transfected with HA-Ubi, Myc-USP3, Flag-EPHA2 or one of four site-specific mutants of EPHA2 (K702R, K778R, K882R and K945R) were treated with 10 µM MG132 for 6 h, and the ubiquitination of the EPHA2 DBD domain was then analyzed.
