A, B MCF10a and C, D 4T1 cells were treated with Tg (0.5 μM) (B, D) or Tm (1.92 μM) (A, C) alone or in combination with MKC8866 (20 μM) for 18 h. Cell lysates were immunoblotted for PERK and XBP1s. E MCF10A cells or F 4T1 cells were treated for the indicated timepoints with Tg (0.5 μM) alone or in combination with MKC8866 (20 μM) and cell lysates immunoblotted for PERK and XBP1s. Actin was used as a loading control. Blots are representative of N = 3. Arrow denotes XBP1s band, * indicates non-specific band(s).
