A Two sgRNA-UBE2M oligos were used to silence UBE2M in MCF7 and T47D cells using the CRISPR/Cas9 system. B–D UBE2M silencing inhibited the growth of both MCF7 and T47D cells. E UBE2M silencing inhibited the colony-forming ability of both MCF7 and T47D cells. F, G UBE2M silencing induced cell cycle arrest. PI staining and FACS analysis were used to analyze the cell cycle profile upon UBE2M silencing (F). Western blotting was used to assess the expression of cell cycle-related proteins, namely, p21, p27 and WEE1 (G). H, I UBE2M silencing induced cell apoptosis.
