a, b Seven-day differentiated Control and SMA human MNs were treated with 20 μM U0126 (U0) during 24 h or left untreated. Cell lysates were then obtained and submitted to western blot analysis using anti-phospho-mTOR (p-mTOR Ser2448), anti-mTOR, anti-p62, anti-SMN, or anti-LC3 antibodies. Membranes were reprobed with anti-α-tubulin antibody (a, b). Graphs represent the expression of p-mTOR or mTOR vs α-tubulin (a), and the expression of p62 or LC3-II vs α-tubulin (b), corresponding to the quantification of at least 3 independent iPSCs differentiation cycles ± SEM. Asterisks indicate differences using Student t-test (*p < 0.05; ***p < 0.001; ****p < 0.0001; ns no significant differences p > 0.05).
