The extrinsic apoptotic pathway is triggered by the binding of the death ligand Fas-L to the transmembrane receptor Fas, which activates caspase-8 and directly clears effector caspase-3/7 to trigger apoptosis execution. Meanwhile, caspase-8-mediated cleavage of BH3-only protein BID generates tBID, which translocate to the mitochondrial surface and cooperate with the pro-apoptotic proteins BAX/BAK to induce MOMP, release Cyt c, and form apoptotic bodies, thereby activating caspase-9-dependent endogenous apoptotic cascade.
