Structural organization of the N-lobe, the C-lobe and the hinge region (indicated with arrows) of the HECT E3s A ITCH, B NEDD4, C WWP1, D WWP2, and E HUWE1. The N-lobe is responsible for interacting with the E2-Ub complex, while the catalytic cysteine (in red), which forms the thioester intermediate, is located within the C-lobe. The hinge region provides the flexibility for conformational changes necessary for the catalytic function.
