a–e The human lung organoids were infected with 323P SARS-CoV-2 or 323L SARS-CoV-2 for 2 h, washed with PBS and replaced with fresh media with 10 μM Nec-1s or vehicle, and incubated for additional 48 h. The cells were lysed with 2× Laemmli sample buffer and analyzed by western blotting using indicated antibodies (a). Total RNA was extracted; viral titers (b) and the expression of ACE2 (c), EGFR (d) and cytokines (e) were quantified by RT-qPCR. Paired t-test was used in qPCR analysis (*P < 0.05, **P < 0.01, ***P < 0.001).
