Overview of the interleukin-1 (IL-1) family system. The IL-1 system comprises seven agonists (IL-1α, IL-1β, IL-18, IL-33, IL-36α, β, and γ), an anti-inflammatory cytokine (IL-37) and eleven receptor molecules (IL-1R1-IL-1R10). Five functional signaling complexes are formed by pairing primary receptors with coreceptors. The system is tightly regulated by negative modulators, including 3 receptor antagonists (IL-1Ra, IL-36Ra, and IL-38), two decoy receptors, IL-1R2 and IL-18 binding protein (IL-18BP), and the inhibitory receptor IL-1R8
