The empty vector (PET101-His) was used as a negative control. The homeodomain transcription factor PtrZFD1 (Potri.002G035200) did not bind to the CAATCAC motif at a gradient of protein concentrations, which further confirmed the binding specificity of PtrWOX13a. The experiments in a and b were repeated independently three times with similar results.
