A Flow cytometry, the gold standard for characterizing cells in bioreactors, requires (1) cell preparation, (2) surface staining with a desired antibody, and (3) sorting and analysis. B An overview of the qOBM integrated imaging system. This includes cells flowing out of the bioreactor through a custom flow cell. The cells are then imaged by the qOBM system illuminated sequentially by four 720 nm LEDs. The cells are then returned to the bioreactor for continued culture. C Healthy, growing T cell culture. D Low viability T cell culture. E Correlation between measured cell density with flow cytometry [Cells/mL] and the qOBM imaged cell density [Cells/mm2]. (F) T cell culture with yeast contamination on day 1 of culture and G 24 h later. H T cell culture with bacterial contamination. Scale bar is 100 μm.
