Mice (n = 10/group) were immunized (i.n.) twice 21 days apart with 1.2 × 1010 v.p. of rAd5-YFV vaccine or rAd5-LcrV vaccine, with animals receiving PBS served as controls (Fig. 7a). Forty-two days after the second vaccination dose or on day 3 post-challenge, spleens were harvested from mice (n = 5/group/timepoint) and stimulated with purified rF1-V (100 µg/mL) for 3 days (b). The cytokines in the culture supernatants were analyzed by using Bioplex-23 assay and expressed as the arithmetic means ± standard deviations. The proinflammatory and anti-proinflammatory cytokines were listed in panel Fig. 7c–g, while the Th1 and Th2 cytokines were displayed in panels h–m, and the chemokines as well as colony stimulating factors are presented in panels n–s. P values were calculated using a two-way ANOVA with Tukey post-hoc test to compare multiple timepoints or student t-test to compare two groups within the same timepoint.
