Processing of AFM images to determine VLP area for the single passed (A–C) and double passed (D–F). The raw AFM images (A and D) were converted to 32-bit grey scale and the edges were enhanced by applying a 2D FFT filter in ImageJ (B and E). After manually adjusting the threshold to maximize the number of fully resolved VLP, the ‘Find Edges’ routine was applied to create a VLP edge map (C and F). Individual VLP area was determined by ImageJ integrating within each numbered contour.
