Reactions containing 100 µM of a model NGLH-containing acyl donor peptide (Ac-RWRGWRNGLH), 200 µM of a Lys-containing acyl acceptor peptide (as indicated in the figure), 200 µM Ni2+, and 1 µM OaAEP1 were run in 100 mM HEPES buffer, pH 8.5, for 3 h at 25 °C. Conversion (%) of the NGLH-containing substrate to isopeptide-linked product was determined via RP-HPLC (n = 3 independent experiments).
