P. falciparum DNA concentration measured by qPCR versus parasitaemia (percentage of infected RBCs) measured by microscopy, for the 87 dried blood spot (DBS) samples collected from Navrongo and sequenced with ONT kit 14 chemistry/ R10.4.1 flow cells in this study. Point size and colour are proportional to the ratio of P. falciparum (Pf) to human DNA concentration inferred by qPCR. Linear regression shows a significant positive relationship between parasitaemia (%) and sample P. falciparum DNA concentration, though with greater variability than for the mock DBS samples (Extended Data Fig. 2). qPCR methods are described in Supplementary Notes.
