a, Representative negative stain EM micrographs and 2D class averages for gB constructs delineate prefusion and postfusion conformations. Approximately 100 micrographs were collected across two independent experiments for each gB construct. b, SPR kinetic binding assays with monoclonal antibodies and the HSV-1 gB co-receptor, PILRa, to compare the antigenicity of prefusion-stabilized and postfusion gB. Antibodies SS10, SS55 and DL16 have been previously reported to recognize discontinuous epitopes dependent on natively folded gB.
