The type I IFN response to cardiac injury begins in the bone marrow

The type I interferon (IFN) response to ischaemic cardiac injury begins in the bone marrow, well before cells infiltrate the injured heart, a recent study has shown. “We previously discovered that innate immune cells in the infarcted heart express an antiviral transcriptional programme called the type I IFN response and that blocking this response improved survival,” remarks senior investigator Kevin King. “Now we show that the IFN response begins far away in the bone marrow and is detectable in the peripheral blood.”

Only one cell population in the heart was found not to have an ISG-expressing subpopulation. Resident macrophages that were in the heart before MI-expressed genes regulated by the transcription factor NRF2 after MI. Mice deficient in NRF2 had increased ISG expression in bone marrow-derived neutrophils and monocytes and monocyte-derived macrophages, suggesting that NRF2 negatively regulates the IFN response to MI.
