CHAPIR, a master regulator of cardiac hypertrophy

Cardiac-hypertrophy-associated piRNA (CHAPIR), a PIWI-interacting RNA (piRNA), accelerates the cardiac hypertrophic response by regulating N6-methyladenosine (m6A) RNA levels.

CHAPIR interacts with PIWIL4, to inhibit the m6A activity of METTL3 (an RNA methyltransferase). Indeed, loss of CHAPIR increased m6A levels, whereas CHAPIR overexpression had the opposite effect. Parp10 was among the genes that were hypomethylated in CHAPIR-overexpressing cardiomyocytes. In CHAPIR knock-out mice, METTL3 binding to Parp10 mRNA was increased in cardiomyocytes, and m6A was enriched in Parp10 mRNA, associated with decreased Parp10 expression.
