Dynamics of antigen retention by follicular dendritic cells

Follicular dendritic cells (FDCs) are cells of stromal origin that are key to the formation of germinal centres (GCs) after infection or immunization. FDCs recruit B cells, regulate their survival and have a crucial role in B cell affinity maturation. FDCs acquire antigens and can display them in their native conformation (as immune complexes) for extended periods of time, which favours affinity maturation and increases antibody specificity. Yet, the dynamics of antigen retention by FDCs remain largely unknown.

In this preprint (not peer-reviewed), Martinez-Riano et al. evaluate the dynamics of antigen retention by FDCs in GCs. The authors immunized mice with fluorescent antigen–immune complexes and followed their localization in draining lymph nodes using 3D confocal microscopy. Whereas the whole FDC network was loaded with antigen 24 hours after immunization, immune complexes accumulated in the centre of the FDC network by day 7. This correlated with greater expression of complement receptor CR2 by central FDCs, which interacts with complement-bound immune complexes. Similar dynamics were also observed using HIV-gp120 nanoparticles, which activate complement without immune complex formation.
