The kiss a cell can’t forget: tracking cell–cell interactions with uLIPSTIC

Cell–cell communication is essential for tissue homeostasis. Although microscopy-based techniques have provided valuable insights into the spatial and temporal regulation of cellular interactions, they have limitations in capturing the transcriptional changes induced by cell contact.

A proximity labelling technique known as LIPSTIC was previously developed to track transient ‘kiss-and-run’ interactions between T cells and dendritic cells (DCs) in vivo. LIPSTIC uses a fusion of the Staphylococcus aureus transpeptidase Sortase A (SrtA) with CD40L, a ligand on T cells, together with an acceptor site, G5, fused to the corresponding receptor, CD40. This allows the enzymatic transfer of a peptide substrate at the CD40–CD40L interface. Although the LIPSTIC approach has led to important discoveries, its applicability is limited by relying on a single receptor–ligand pair.
