Oral tolerance is a fundamentally important mechanism for our immunological coexistence with commensal microbiota and dietary antigens. Failure to establish such tolerance promotes food allergies and chronic inflammation. RORγt⁺ antigen-presenting cells (APCs), including dendritic cell-like Thetis cells (TCs) and group 3 innate lymphoid cells (ILC3s), are important in establishing oral tolerance. However, the molecular mechanisms that control RORγt expression within these APCs remain unclear.

In this preprint (not peer reviewed), Guo et al. identify the open chromatin region 369 (OCR369) as essential for the development of RORγt+ APCs, including RORγt⁺ ILC3 and TC subsets. Through in silico analysis of publicly available ATAC-seq data, the authors find that OCR369 is uniquely accessible in RORγt⁺ APCs and ILC3s but not in RORγt+ T helper 17 (TH17) cells.
