a, Hamming distance was calculated between the 11 specificity residues of each PhoQ*–PhoP* variant pair, and the 11 specificity residues of wild-type PhoQ–PhoP, the two-component signalling paralogues in E. coli or all extant two-component signalling proteins. When comparing two sets, all distances between all members of both sets are plotted. b, Force-directed graph representing the sequence space of histidine kinases.
