The maximal response is reported as a percentage of the maximum effect at the WT. Data are mean ± s.e.m. from at least three independent experiments performed in technical triplicate. NA, not active. Expression levels of PTH1R mutants. The relative expression levels of PTH1R in AD293 cells were determined by flow cytometry (FACS) using WT as the reference. Cells were gated with a forward scatter cutoff at a gain value of 0.18% by using the blank sample as the reference value of background fluorescence signal. The left region represents the negative cell community, while the right region represents the positive cells that express the receptor.
