(a) Representative flow cytometry plots showing time courses of apoptosis following drug activation of StaPLd-Casp9 or iCasp9. Parallel time courses were also performed with DMSO vehicle control to account for any cell death due to nutrient depletion over time. RFP signal serves as an indicator for suicide gene expression. Cells were stained and fixed for flow cytometry analysis after indicated time in drug. (b) Time course of cell death induced by activation of StaPLd-Casp9 or iCasp9. StaPLd-Casp9 takes 24 h to reach near-maximal apoptosis, whereas iCasp9 does so by 3 h. Mean ± s.e.m. are plotted. Dots represent values from 3 independent experiments.
