The excitation laser (blue beam) emerges from the lower right, is reflected by the dichroic mirror (DM1), is then split by the non-polarizing beamsplitter (NPBS), and finally interferes at the sample plane. The emitted fluorescence (green beam) undergoes interference at the NPBS before being captured by the sCMOS camera (CAM1). The focus-lock laser (red beam) emerges from the upper left, is reflected by the dichroic mirror (DM2), is directed toward the objectives (OBJ1, 2), and is finally captured by a compact CMOS camera (CAM2). The excitation laser for I2M illumination (dark blue) emerges from the lower left and is reflected by a dichroic mirror (DM3) to the sample.
