(a) Two bar graphs detailing the localization patterns of C18orf21-GFP, RPP21-GFP, and RPP14-GFP in HeLa cells. The graph on the left shows the percentage of cells that contained nuclear puncta. Nuclear puncta were defined as GFP signal greater than 0.25 µm in size and with an integrated pixel intensity above background. The graph on the right shows the percentage of cells that contain an enriched nucleolar GFP signal. The number of cells counted for each condition were: C18orf21 n=86, RPP14 n=39, and RPP21 n=34 cells. (b) Representative co-immunofluorescence images from two biological replicates of cell lines that ectopically express the indicated C-terminally GFP-tagged construct. Prior work suggested that RNase P localizes to Cajal bodies36,80. To mark Cajal bodies we co-stained each cell line for Coilin using an anti-coilin antibody (see materials and methods). The bottom inset depicts a magnified view of the merged GFP and Coilin staining. Here we find that C-terminally tagged constructs are not found in Cajal bodies suggesting that RNase P does not localize to these structures.
