The differential effects of LPS are independent of IL-6 activity. (A) The supernatants from monocytes at 0, 3, 6 and 24 hours post LPS (500 ng/ml) stimulation were harvested and analysed for IL-6 production by ELISA n = 2. (B) The supernatants from uninfected monocytes (mono) or long term latently infected monocytes (Mono + HCMV) at 0, 3, 6 and 24 hours post LPS (500 ng/ml) stimulation were harvested and analysed for IL-6 production by ELISA n = 2. (C) Monocytes were stimulated with 500 ng/ml LPS, 50 ug/ml IL6, 500 ng/ml LPS plus nIL6 antibody, or 50 ug/ml soluble IL6 receptor (sIL6r) for 24 hours then infected with Merlin and then stained for IE protein expression 24 hpi. Nuclei were counter-stained with DAPI and infection rate calculated n = 3. **p < 0.01; NS = non-significant difference when compared to infection of LPS control.
