Omission of an SDS denaturation step. Omitting the additional denaturation step, which uses heating to 60 °C for 10 min in 2% SDS, did not change the EPO N-glycan profile. This is beneficial, as SDS may interfere with PNGaseF release and MALDI analysis. Additionally, incubation at elevated temperatures bears the risk of desialylation.
