CoQ0 triggers apoptotic DNA fragmentation and promote death of SKOV-3 cells. (A–D) Cells were pretreated with caspase inhibitor (Z-VAD-FMK, 20 μM) for 1 h followed by CoQ0 (0–30 µM) for 24 h. (A) Apoptotic DNA fragmentation was determined by TUNEL assay. The green florescence indicates TUNEL-positive cells in the microscopic fields (200 × magnification) from three separate samples. (B) The fold of apoptotic cells was calculated by quantifying the florescence intensity using commercially available software. (C) Cleavage of PARP was estimated by Western blot. Changes in protein intensities were quantified by commercially available software. (D) Cell viability with or without Z-VAD-FMK treatment was determined by MTT assay. Values expressed as mean ± SD of three independent assays (n = 3).
