E1, L1 and L2 bind endogenous pMHC targets on EBV-transformed BLCLs. (A,B) T2 cells were pulsed with 10-fold serially diluted peptides before staining with (A) murine IgG1 or (B) chimerized IgG1 TCR-like mAbs. Cells were then stained with AlexaFluor-488 conjugated secondary antibodies for detection via flow cytometry. (C) Representative histogram plots of A*02:01, A*02:03, A*02:06 and A*02:07 EBV BLCLs stained with the respective chimeric TCR-like mAbs to detect endogenously presented pMHC surface targets. A*11:01 and A*24:02 BLCLs were used as controls. (D) Normalized mean fluorescence intensity (nMFI) of EBV BLCLs stained with the three TCR-like mAbs. Values were normalized by dividing the MFI reading from that of their respective secondary antibody control staining.
