EBr induced nucleosome destabilization. (A) EBr induced elution of H2B-GFP and H3-GFP performed on ice and at room temperature (RT) in histone-GFP expressor HeLa nuclei. EBr was applied in the presence of 0.75 M NaCl. (B) Effect of co-treatment with EBr and salt in histone-GFP expressor HeLa nuclei. The shift of elution curves in the presence of 100 µg/ml EBr is seen comparing the continuous and dashed lines. The blue double arrow shows that the maximal effect of EBr on nucleosome stability occurs at 0.75 M salt. (C) EBr elution profiles of H3K4me3 or H3K27me3 in HeLa nuclei.
