Mutations of R81 and R88 promote formation of disassembly intermediates in IβIα nucleosomes. (a) Normalized spFRET histograms for wild type and RA-mutants (top), and for wild type and RE-mutants (bottom) at 400 mM NaCl. From each histogram the relative proportion of each of the three subpopulations (LF, MF, HF) was calculated from the areas multi Gaussian fit. (b) Percentage of partially disassembled nucleosomes (HF) relative to the sum of intact plus partially disassembled nucleosomes (MF + HF), elucidating correlations between enhancement of the HF state and decrease in nucleosome stability provoked by charge-modifying mutations in the H2A α3 domain. Averaged proportions and standard error of the mean were calculated from three independent replicas.
