From: Rab5 is critical for SNAP23 regulated granule-granule fusion during compound exocytosis

Inhibition of IKKβ2 prevents SNAP23 promoted formation of super giant SGs in IgE/Ag triggered cells. RBL cells were triple-transfected with 10 μg NPY-mRFP, 15 μg of pEGFP-CA Rab5A and 20 μg of HA-wt-SNAP23. Cells were either left untreated (UT) or sensitized with 1 μg/ml of IgE. Twenty-four hours after transfection, cells were pre incubated with BMS-345541 (5 μM) or vehicle and further incubated for 10 min without or with 50 ng/ml DNP-HSA (Ag) as indicated. Cells were fixed and immunostained using monoclonal antibodies directed against HA followed by Hilyte Plus 647-conjugated goat anti-mouse IgG. Cells were analyzed by confocal microscopy. Bars = 5 μm.
