Results of quantitative RT-PCR. To confirm the effect of TNF-α on osteogenic gene expression in C2C12 cells, the cells were incubated for 0, 4, 8 and 48 h in the presence of various concentration of TNF-α (0.1 to 10 ng/ml). At 4 h, an increase in ALP mRNA expression level was observed when the cells were treated with low concentrations of TNF-α (0.1 and 1 ng/ml), in contrast to the results for 10 ng of TNF-α treatment. The levels of Runx2 mRNA expression increased from 8 h when the cells were treated with low concentrations of TNF-α (0.1 and 1 ng/ml). In the case of osteocalcin, the level of mRNA expression was not significantly different among groups. At 48 h, the level of ALP, osterix mRNA expression was increased in 1 ng/ml of TNF-α treatment.
