SUMOylation may regulate chromosome behaviors via BmPlk1 during mitosis. (a and b) RNAi of ubiquitylation genes and SUMOylation genes induced abnormal mitotic spindle in cultured silkworm cells. The mitotic spindle was stained with anti-Tubulin antibody (green) and nucleus DNA was visualized by DAPI (blue). Arrows indicated the misaligned and lagging DNA. Scale bar, 10 µm. (c) Silkworm cells stably expressing the histone protein EGFP-H4 were treated with indicated dsRNAs and analyzed by live-cell microscopy. RNAi of BmSMT3/BmUBC9 or BmPLK1 resulted in the failure of chromosome congression and segregation, even after a prolonged duration of mitotic delay. Scale bar, 10 µm. The percentages of silkworm cells harboring the abnormal progression were calculated in selected mitotic cells.
