Analytical ultracentrifugation reveals direct binding of exportin-1 and importin α2/β1 to HeV V. Sedimentation velocity analytical ultracentrifugation experiments were performed on purified recombinant exportin-1/Ran-GTP, importin α2/β1, wild-type HeV V and variants thereof, both alone and in various combinations. The continuous sedimentation coefficient distribution [(c)s] was plotted as a function of s20,w (black) for HeV V (a,b) wild-type, (c,d) VL174A/L177A, (e,f) V1-406, and (g,h) V51-457. Exportin-1/Ran-GTP (blue) and importin α2/β1 (green) are shown. Equimolar V:exportin-1/Ran-GTP mixtures are shown in magenta and V:importin α2/β1 mixtures are shown in orange.
